0012 - Alendronate Therapy Increases the Mechanical Properties of Canine Trabecular Bone
ثبت نشده
چکیده
Introduction: Alendronate, a second generation bisphosphonate, is known to increase bone density by inhibiting osteoclastic bone resorption. It is clincally used to treat osteoporosis, Paget’s disease and hypercalcemia associated with malignancy [1]. In clinical trials and animal studies, the effects are typically quantified by measuring bone mineral density. In this study, we sought to determine whether healthy canine bones exhibit increased structural or mechanical properties following alendronate treatment. We tested both whole long bones and trabecular bone samples, because a prior study with pamidronate had different findings with these two sample types [2]. We hypothesized that alendronate treatment would increase the structural properties of bones and mechanical properties of bone tissue in healthy dogs.

Comparative effects of alendronate and alfacalcidol on cancellous and cortical bone mass and bone mechanical properties in ovariectomized rats.
The purpose of the present study was to compare the effects of alendronate and alfacalcidol on cancellous and cortical bone mass and bone mechanical properties in ovariectomized rats. Twenty-six female Sprague-Dawley rats, 7 months of age, were randomized by the stratified weight method into four groups: the sham-operated control (Sham) group and the three ovariectomy (OVX) groups, namely, OVX ...

current study were to develop a mathematical model that simulates control and 1 year experimental results following bisphosphonate treatment (alendronate or risedronate) in the canine fourth lumbar vertebral body, validate the model by comparing simulation predictions to 3 year experimental results, and then use the model to predict potential long term effects of bisphosphonates on remodeling a...

One of the main limitations of glucocorticoid therapy is its negative effects on bones. The aim of this study was to determine the effects of concurrent administration of eicosapentaenoic acid (EPA) and alendronate on bone changes induced by glucocorticoid treatment in rats. Thirty six male Wistar rats, who were 2.5 months of age, were divided equally into six groups and treated with normal sal...

Bisphosphonates suppress bone remodeling activity, increase bone volume, and significantly reduce fracture risk in individuals with osteoporosis and other metabolic bone diseases. The objectives of the current study were to develop a mathematical model that simulates control and 1 year experimental results following bisphosphonate treatment (alendronate or risedronate) in the canine fourth lumb...

One year of treatment with bisphosphonates at 5x the dose used for post-menopausal osteoporosis significantly increases failure load and microdamage, and decreases toughness at multiple skeletal sites in intact female beagles. The goal of this study was to determine if similar changes occur with doses equivalent to those used for post-menopausal osteoporosis treatment. Skeletally-mature female ...
